Abstract:
 Cellulase and xylanase production by Leucoagaricus meleagris KKU-C1 isolated from compost at pulp and paper industry, was studied in solid state fermentation using agricultural wastes as substrates. The highest cellulase and xylanase activity was obtained using a mixture of rice bran and rice husk as carbon source and sodium nitrate as nitrogen source, respectively. Crude carboxymethyl cellulase (CMCase) exhibited the highest activity at pH 8.0 and 40 ˚C, whereas crude xylanase did at pH 8.0 and 50 ˚C, respectively. Crude CMCase and crude xylanase retained over 46% and 52% of the original activity in the pH range of 7.0-11.0, respectively, after incubation at 4 ˚C for 60 min. As for thermo-stability, crude CMCase and crude xylanase retained over 50% and 69% of the original activity up to 50 ˚C, respectively, after treated at pH 8.0 for 60 min. Thus, crude CMCase and xylanase from L. meleagris KKU-C1 exhibited the highest activity at pH 8.0 and stable in alkaline pH region, suggesting that these enzymes could be used in the pulp industry.
