Abstract: Mycelia of the Ling-Zhi mushroom cultivar L6 (Ganoderma lucidum) were cultured for 28 days in the medium containing 2% (by volume) molasses and 2% (by weight) soybean flour at room temperature on rotary shaker at 150 rpm. The glycosidase activities were assayed both in the supernatant of the medium and within mycelia extracted with PBS by hydrolysis of specific p-nitrophenyl glycosides. Total activities of the extra- and intracellular glycosidases were more or less the same and total protein of the intracellular fractions was about 1.5 fold of the extra-ones. The extracellular fraction contained 217 mU/ml
a -galactosidase, 41 mU/ml b -glucosidase and less than 11 mU/ml of b -galactosidase, a -glucosidase, b -xylosidase and b -N-acetylhexosaminidases. However, the intracellular fraction contained 198 mU/ml b -glucosidase and 44 mU/ml a -galactosidase. During 28 days of the cultivation peroid, the intracellular glycosidase activities were increased, stable and then decreased while the extra-ones were increased and stable. All of the results suggested that the glycosidases were synthesized, accumulated in the mycelium and then selectively excreted into the medium. |
